CAR-T therapy harnesses the body’s own T cells to target and destroy cancer cells. In this approach, a patient’s T cells are engineered to express a chimeric antigen receptor (CAR) that recognizes specific antigens on cancer cells, enabling precise killing. Recent breakthroughs, such as the use of deep learning to design binding proteins for peptide–MHC-I complexes, are opening new pathways for even more targeted immunotherapies.
To develop effective CAR-T treatments, identifying high-affinity binding domains for cancer antigens is essential. While traditional methods rely on animal immunization and antibody screening, peptide screening offers a faster, animal-free alternative.
Peptide Library Screening – A Streamlined Workflow
We support CAR-T and therapeutic discovery with ready-to-use and custom peptide library kits (e.g., random 7-mer, 12-mer, cyclic 7-mer) using phage display technology. Our process includes:
Library Construction
Random peptide sequences are cloned into phage coat proteins (e.g., M13 pIII/pVIII), creating a physical link between the displayed peptide and its DNA.
Affinity Screening (Biopanning)
Binding
Target molecules are immobilized, and the phage library is introduced.
Washing
Weakly bound phages are removed.
Elution
Specifically bound phages are recovered.
Amplification
Eluted phages are amplified in E. coli for enrichment.
Iterative Rounds
Typically 3–5 rounds are performed to enrich high-affinity binders.
Identification & Analysis
Binding Validation
Positive clones are confirmed via phage ELISA.
DNA Sequencing
Peptide sequences are determined.
Affinity Analysis
Binding strength is measured using SPR, ELISA, or similar.
